Molecular and Cellular Pathobiology Myc Posttranscriptionally Induces HIF1 Protein and Target Gene Expression in Normal and Cancer Cells
نویسندگان
چکیده
c-Myc is frequently overexpressed in tumors and plays an important role in the regulation of cancer metabolism. Hypoxia-inducible factor-1 (HIF1), the master regulator of the hypoxic response, enhances tumorigenesis and influences metabolism via upregulation of the glycolytic pathway and suppression of mitochondrial respiration. Together, deregulated Myc and HIF1 cooperate to lend metabolic advantages to proliferating cancer cells and contribute to the Warburg effect. Here we show that overexpression of Myc significantly stabilizes the a subunit of HIF1 (HIF1a) under normoxic conditions and enhances HIF1a accumulation under hypoxic conditions in cells. Posttranscriptional regulation of HIF1a by Myc led to the induction of HIF1a gene targets. Normoxic HIF1a protein expression was also dependent on Myc. Functionally, HIF1a expression was required for Myc-induced anchorage-independent growth and cell proliferation. Mycdependent stabilization of HIF1a involved either disruption of binding to the VHL complex or posttranslational proteinmodifications. Taken together, our findings uncover a previously uncharacterized regulatory relationship between Myc and HIF1 that has important implications for cancer metabolism and development. متن کاملStudy of NGEP expression in androgen sensitive prostate cancer cells: A potential target for immunotherapy
Background: Prostate cancer is one of the leading causes of cancer deaths among men. New gene expressed in prostate (NGEP), is a prostate-specific gene expressed only in normal prostate and prostate cancer tissue. Because of its selective expression in prostate cancer cell surface, NGEP is a potential immunotherapeutic target. To target the NGEP in prostate cancer, it is essential to investig...
متن کاملGenistein Induces Apoptosis and Inhibits Proliferation of HT29 Colon Cancer Cells
Soybean isoflavone genistein has multiple anticancer properties and its pro-apoptotic and anti-proliferative effects have been studied in different cancer cells. However, the mechanisms of action of genistein and its molecular targets on human colon cells have not been fully elucidated. Therefore, caspase-3 and p38 mitogen-activated protein kinase (p38 MAPK) as the main therapeutic targets...
متن کاملEffect of valproic acid on JAK/STAT pathway, SOCS1, SOCS3, Bcl-xL, c-Myc, and Mcl-1 gene expression, cell growth inhibition and apoptosis induction in human colon cancer HT29 cell line.
Background and aim: Cytokines are a large family of protein messengers. These proteins induce various cellular responses. Janus kinases (JAKs) are mediators of cytokine, activated JAKs phosphorylate signal transducers, and activators of transcription (STAT) proteins that regulate cell differentiation, proliferation, and apoptosis. Aberrant JAK/STAT signaling is involved in the oncogenesis of se...
متن کاملA Mimic of the Tumor Microenvironment on GPR30 Gene Expression in Breast Cancer
Introduction: The G-protein coupled receptor 30 (GPR30) gene is a member of the G-protein coupled receptor (GPCR) family; involved in breast, endometrial, and ovarian cancers. Many GPCR receptors that are implicated in several types of human cancers are correlated with increased cell proliferation and tumor progression; especially GPR30 gene. Methods: The breast cancer MCF-7 and MDA-MB-231 cel...
